Abstract

 Halophiles are microorganisms that inhabit environments with high salt concentrations. In recent years, there has been growing interest in studying these microorganisms due to their biotechnological potential, particularly in the production of enzymes with industrial relevance. Samples were collected at Bahía de Santa Barbara, Sonora, Mexico. A total of 287 colonies were grown on Marine agar 1217 and ATCC 2185 agar at NaCl concentrations of 0.5, 2 and 4 M, 35 colonies showed hydrolysis halos on 0.5 M marine medium, 7 on 2 M marine medium, and 4 on 2 M ATCC 2185 medium, all of them supplemented with 2% skim milk as a proteolytic activity inducer. Five strains (5.3.3, 5.3.4, 5.3.6, 5.3.8, and 4.2.11) were selected based on the size of the diameter of the hydrolysis halo observed on the inducer-containing medium. Protein concentration analysis indicated that strain 5.3.8 exhibited the highest concentration (144.46 µg/mL) since the beginning of fermentation. Proteolytic activity was detected specifically after 144 hours of fermentation, where strain 5.3.3 was the one showing the highest proteolytic activity. Enzymatic activity levels were 0.2875, 0.0402, 0.2201, 0.2323, and 0.2035 U/mL for strains 5.3.3, 5.3.4, 5.3.6, 5.3.8, and 4.2.11, respectively. These results suggest that slightly halophilic strains from saline soils were isolated representing a promising source of proteolytic enzymes with biotechnological potential.
